Job Responsibilities

Molecular Research Lab (MRL) harbours a multi-disciplinary research platform in a state-of-the-art fashion provided with a good number of cutting edge analytical tools. I am involved in both DNA lab and Proteomics division of MRL. My key responsibility is to develop new clones for optimizing existing process, lab scale expression and purification of desired proteins from new clones, master cell bank characterization, 16S rRNA based characterization of unknown microbes found in the facility, mass spectrometric characterization of protein drugs.

Research Activities

Development of cost effective Pichia pastoris clone secreting Insulin Glargine: In our previous strategy our secretory protein was des 30 Insulin Precursor (have no B30 Threonine). To add the B30 Thr in downstream we have to use H-Thr(tBu)-OtBu ester, a high cost reagent that affect the COG of the final product. To overcome the etherification we design our gene construct so that it secretes full Insulin Glargine where A and B chain is connected with short linker peptide and removed during downstream processing.

Protein Characterization: Proteins’ primary characterizations – Intact molecular mass detection, peptide mapping, amino-acid sequencing (De-Novo sequencing), top-down analysis of intact protein, impurities profiling (identification of process variants and post-translational modifications) analysis using high resolution ESI-TOF (Triple TOF 5600, AB Sciex).

Protein Purification: Purification of proteins using gel filtration, ion exchange, affinity and hydrophobic interaction chromatographic (HIC) methods.

Two-step Transpeptidation: This method is the yield-limiting factor in the recombinant production of human-Insulin in Pichia Pastoris, which includes the peptide addition of Threonine at B-29, followed by enzymatic digestion of Insulin Precursor (IP). We increased the yield of the final product nearly two-fold by segregation of the method into two steps. In the first step, we tuned the enzymatic digestion and then Threonine addition followed by a chromatographic purification.

Mass spectrometric characterization of Salmonella typhi membrane proteins: Purification and identification of antigenic membrane protein of Salmonella typhi following chromatographic (gel filtration) purification then In-gel digestion, tandem mass spectrometric analysis (bottom-up).
